绿原酸对D-半乳糖诱导的大鼠认知功能障碍的保护作用及机制研究
Study on the protective effect and mechanisms of chlorogenic acid against D-galactose-induced cognitive dysfunction in rats

Abstract
In order to investigate the protective effect of chlorogenic acid(CGA)against D-galactose induced cognitive dysfunction in rats and its underlying mechanism,Sprague-Dawley(SD)rats were treated with D-galactose to induce aging-related cognitive decline.Interventions were designed for low and high-dose chlorogenic acid groups.The memory ability was evaluated by Morris water maze test,the pathological morphology of hippocampus was examined by HE staining.Oxidative stress markers(the levels of MDA,SOD,GSH-Px,CAT)in rats brain tissue were analyzed using kits,and inflammatory cytokines(the levels of IL-1β,IL-6,TNF-α)in rats serum were measured using ELISA.Furthermore,the expression of Nrf2,HO-1 and NQO1 proteins was evaluated by Western blot.It was found that CGA at both low and high doses improved cognitive performance,as evidenced by reduced escape latency(P<0.01)and increased platform crossings frequency and target quadrant occupancy(P<0.01).It also improved hippocampus histology,enhanced enzyme activities of SOD,GSH-Px and CAT in rats brain tissue,and reduced accumulation of MDA(P<0.01).The expression levels of IL-1β,IL-6 and TNF-α were decreased(P<0.01),and expression levels of Nrf 2,HO-1 and NQO-1 were increased(P<0.01).These findings suggest that chlorogenic acid can improve learning and memory deficits induced by D-galactose in rats to some extent,and its possible mechanism is associated with the enhancement of antioxidant capacity and the reduction of inflammatory responses.关键词
