Abstract

A single-factor experimental design was employed to study the extraction of total flavonoids from Codonopsis pilosula(C-Fla).The effect of ultrasonic temperature,ultrasonic time,ethanol concentration,solid-to-liquid ratio and extraction pH on total flavonoid extraction yield was studied.Response surface methodology was used to design the pairwise interaction response surface of three factors with the total flavonoid yield as the optimal index.Following purification with macroporous resin,the anti-hair loss efficacy of the extracted total flavonoids was evaluated.The optimal extraction conditions were found to be as follows:ultrasonic temperature of 48℃,ultrasonic time of 90 minutes,ethanol concentration of 84%,solid-to-liquid ratio of 1∶20 g/mL,and extraction pH of 7.Under these conditions,the flavonoids extracted yield reached 21.680 mg/g.Furthermore,the extracted flavonoids exhibited excellent antioxidant activities,effectively inhibited 5α-reductase activity and promoted cell repair in a DHT-induced alopecia model.These results showed that Codonopsis pilosula flavonoids hold considerable potential in promoting hair growth.The study provides a theoretical basis for further development of Codonopsis pilosula resources.
