Abstract
[Objective]To establish an ultra-performance liquid chromatography-atmospheric pressure chemical ionization-tandem mass spectrometry(UPLC-APCI-MS/MS)for the determination of stigmasterol,campesterol,β-sitosterol and stigmastanol by multi-reaction monitoring(MRM)mode in the sea asparagus.[Method]Using ACQUITY UPLC BEH C18 column(100 mm×2.1mm,1.7 µm),0.1%formic acid aqueous solution-methanol as the mobile phase,with a flow rate of 0.5 mL/min.Using atmospheric pressure chemical ion source(APCI),and selected multi-reaction ion monitoring(MRM)detection mode.[Result]Totally 4 sterols had good linear relationships in the linear ranges of 2-1 000 µg/kg and 5-1 000 µg/kg,and the recoveries were between 92%—112%.The LODs of stigmasterol,campesterol,β-sitosterol,stigmastanol were 5 µg/kg,2 µg/kg,2 µg/kg,5 µg/kg,and their LOQs were 10 µg/kg,5 µg/kg,5 µg/kg,10 µg/kg respectively.[Conclusion]Due to its high sensitivity and fast analysis speed,this method has been successfully applied to the analysis of sterol content in different harvest stages of sea asparagus,which provides a reference for the determination of other phytosterol content.关键词
