基于网络药理学探讨黄精治疗糖尿病的活性成分及作用机制
Exploring The Potential Active Components and Mechanism of Action of Polygonati rhizoma in Treatment of Diabetes Mellitus:Based on Network Pharmacology

Abstract
[Objective]To explore the potential active component and mechanism of action of Polygonati rhizoma for the treatment of diabetes mellitus using network pharmacology and molecular docking technology.[Method]The chemical composition,target of action and disease-related target of the drug were obtained from TCMSP herbal system pharmacology platform and DisGeNET,NCB and TTD databases,respectively.Cytoscape 3.8.0 software was used for the construction of drug-active ingredient-target-of-action(DOA-TOA)network diagrams.String database was used for the construction of The PPI network model was constructed using String database.GO and KEGG functional analyses were performed using Metascape platform;molecular docking was performed using Pubchem database,PDB database,Chem Bio3D Ultra 14.0 software,PyMOL 2.4.0 software and Auto Dock Tools 1.5.6 software.[Result]A total of 8 active components and 75 targets were screened,and 29 targets were shared by Polygonati rhizoma and diabetes mellitus.The main active components were DFV,β-sitosterol,diosgenin and baicalein,etc.The core targets obtained from the PPI network were AKT1,TP53,HIF1A,etc.KEGG was enriched with a number of targets for cancer,hepatitis,lipid and atherosclerosis and other multiple signaling pathways.Molecular docking results showed that the key active ingredients and the core targets were free to bind,and β-sitosterol had a strong interaction with AKT1.[Conclusion]The treatment of diabetes by yellow essence is characterized by multi-components,multi-targets and multi-pathways,and its mechanism of action may be related to multiple signaling pathways such as cancer,hepatitis,lipid and atherosclerosis.关键词
