Optimization of the extraction technology of volatile oil from Qinling Artemisia argyi by ultrasound combined with enzyme-assisted distillation and its antioxidant activity

Abstract

Based on a single-factor experiments,under the conditions of a fixed ultrasonic power of 100 W,a solid-to-liquid ratio of(1∶20)g/mL and an enzymatic hydrolysis time of 60 min,the response surface method was used to optimize the extraction process of volatile oil from Qinling Artemisia argyi by ultrasonic combined with enzyme-assisted distillation,and the antioxidant activity was assessed by scavenging DPPH and ABTS+free radicals.The results showed that when the extraction time was 4.3 h,the enzymatic hydrolysis temperature was 54℃,the enzyme amount was 125 U/g,and the ultrasonic time was 47 min,the extraction rate of volatile oil from Qinling Artemisia argyi was 0.7549%,which was 0.0749%lower than the predicted value by the software.The results of antioxidant test showed that when the concentration of volatile oil was 1.0 mg/mL,the scavenging rates of DPPH and ABTS+free radicals were 91.83%and 92.53%,respectively,which were close to the scavenging rates of positive control Vc at the same concentration(94.48%and 95.32%),but there was no significant difference(P>0.05).The clearance rate increased with the in-crease of concentration.This study provides a reference for the development and utilization of Artemisia argyi resources in Qinling Mountains.
