Abstract

This study investigated the therapeutic effects of the novel formulation PURCUMIN® curcumin FC-11 on knee osteoarthritis(KOA)in rats.Firstly,the stability of FC-11 was determined by high-speed centrifugation,analysis of color difference,particle size measurement,Zeta potential assessment,and instant solubility tests,and then the anti-inflammatory characteristics of FC-11 were verified.SD Male rats were randomly divided into a normal group,model group,diclofenac sodium group(5 mg/kg),curcumin groups(20、40、80 mg/kg)and a combined treatment group of curcumin and glucosamine.Except the normal group,the other groups were induced by sodium iodate to establish rat KOA model.The mechanical pain threshold was measured by up-and-down method,the pain latency was measured by dry ice refrigeration method,and the joint swelling was measured by thread binding method.The articular cartilage tissue was evaluated by hematoxylin-eosin and Fano-fast green staining and Mankin's score,and the spleen was weighed to calculate the spleen index.The number of white blood cells was detected by blood cell analyzer,and the levels of inflammatory mediators TNF-α,IL-1β and PGE2 in serum were detected by Western blot.The results showed that both the novel formulation curcumin FC-11 and the combination of FC-11 and aminoglycans significantly improved the inflammatory symptoms associated with rats' knee osteoarthritis.
