南阳育龄妇女MTHFR和MTRR基因多态性及与血清叶酸、同型半胱氨酸、维生素B12的相关性分析
Study on Distribution of MTHFR and MTRR Gene Polymorphisms and Their Relationship with Blood Folate,Homocysteine and B12 in Women of Childbearing Age in Nanyang

Abstract
[Objective]To investigate the genetic polymorphisms of methylenetetrahydrofolate reductase(MTHFR)C677T,A1298C and methionine synthetase reductase(MTRR)A66G in women of childbearing age in Nanyang,Henan Province,and to analyze the relationship between different genetic characteristics and serum folate,homocysteine and vitamin B12.[Method]A total of 122 healthy women of childbearing age in Nanyang,Henan Province were recruited.Saliva was collected for gene polymorphism analysis of MTHFR C677T,A1298C and MTRR A66G.Blood was collected to detect serum folate,homocysteine and vitamin B12.[Result]The genotype frequencies of MTHFR C677T CC,CT and TT were 17.2%,44.3%and 38.5%,respectively;the genotype frequencies of MTHFR A1298C AA,AC and CC were 80.3%,19.7%and 0.0%,respectively;and the genotype frequencies of MTRR A66G AA,AG and GG were 49.2%,43.4%and 7.4%,respectively.The serum folate levels of women with MTHFR C677T CT and TT genotype were significantly lower than those with CC genotype(P<0.05).No significant difference was observed in serum folate,homocysteine and vitamin B12 in women with different MTHFR A1298C or MTRR A66G genotypes(P>0.05).There was no significant difference in serum folate,homocysteine and vitamin B12 among women with different folate utilization ability(P>0.05).There was a linear negative correla-tion between folate and homocysteine in serum of women,with a correlation coefficient of-0.310.[Conclusion]The genotype frequencies of MTHFR C677T,A1298C and MTRR A66G in women of childbearing age in Nanyang were basically the same as those in northern China.In this test,the risk level of folic acid utilization ability is not consistent with the level of serum folic acid and homocysteine,and the results need to be verified in large population studies.关键词
