Optimization of extraction process of tea polyphenols from Liupao tea using deep eutectic solvent method and their antioxidant activity

Abstract

Ultrasonic assisted deep eutectic solvent(DES)was used to extract tea polyphenols from Liupao tea,and the extraction process was optimized based on single factor experiments combined with response surface analysis.The antioxidant capacity of Liupao tea polyphenols was explored.The results showed that the yield of extracting polyphenols from Liupao tea using urea-choline chloride based DES was better than that obtained using anhydrous ethanol;The optimal extraction process conditions for tea polyphenols in Liupao tea were as follows:an extraction time of 21.63 min,DES water content of 31.78%,a material to liquid ratio of 113 mL/g.Under the conditions above,the predicted yield of Liupao tea polyphenols was 14.04%.In vitro antioxidant experiments showed that the IC50 values of Liupao tea polyphenols for DPPH·,·OH,and·O2-scavenging rates were 3.9,1.19,0.19 mg/mL,respectively,indicating that Liupao tea polyphenols had antioxidant activity.
