Identification of bitter substances from Allii tuberosi semen based on UPLC-Q-TOF-MS/MS and their immunoregulatory activity

Abstract
In order to clarify the sources of bitter components in Allii tuberosi semen,column separation and chromatographic separation techniques were used to isolate,purify bitter substances from allii tuberosi semen extract.Then,ultra performance liquid chromatography-quadrupole-time-of-flight mass spectrometry(UPLC-Q-TOF-MS/MS)combined with sensory evaluation was used to identify the obtained bitter components from Allii tuberosi semen.The results showed that the bitter substances in Allii tuberosi semen were mainly thymine,guanosine,and trigonelline.Compared to the Allii tuberosi semen extract obtained by ethanol extraction,the bitter values of the Allii tuberosi semen extract obtained by aqueous extraction were higher.The zebrafish model was used to evaluate the immunomodulatory activity of bitter components from Allii tuberosi semen extract.The bitter component trigonelline in Allii tuberosi semen extract demonstrated significant enhancement of macrophage phagocytosis and alleviation of T-cell depletion in zebrafish at low and medium doses.The research results provide data support for targeted bitterness reduction in Allii tuberosi semen extract while preserving their immunomodulatory activity.关键词
