Cloning and Low-Temperature Stress Response of VabHLH123 Gene in Vitis Amurensis

Abstract
This study cloned the basic helix-loop-helix gene VabHLH123 from Vitis amurensis to investigate its role in cold stress response,providing a theoretical basis for research on the function of low-temperature resistance genes in grapevine.The expression and function of VabHLH123 were investigated through bioinformatic analysis,subcellular localization,low-temperature and exogenous hormone treatment,as well as transient expression.The coding sequence of the VabHLH123 gene is 279 bp,encoding 92 amino acids and containing an HLH domain.The tissue-specific expression analysis showed that VabHLH123 was expressed in all tissues,with the highest relative expression level in the stem.The expression level of VabHLH123 was upregulated after cold stress treatment.After GA3 treatment,the VabHLH123 gene responded rapidly and reached its highest expression after 1 hour,showing an overall upregulation trend.After ABA treatment,the VabHLH123 gene expression was at its highest after 6 hours.Treatments with MeJA,ETH,and SA induced only slight responses in VabHLH123 expression.The subcellular localization results revealed that VabHLH123 is localized in the nucleus.The transient transformation of grape callus showed that cold resistance genes(VvCBF1,VvCBF3,VvRD29B and VvNCED1)had higher expression in VabHLH123 overexpression callus under cold stress.Transient overexpression of VabHLH123 significantly increased the expression level of cold-related genes and enhanced cold tolerance of grape callus.关键词
