Optimization of extraction process of total polyphenols and total flavonoids from different parts of black chokeberry and their antioxidant activities

摘要

Abstract

Black chokeberry was used as the raw material,and total polyphenols and total flavonoids contents in its different parts(roots,stems,leaves,fruits)were measured by Folin-Ciocalteu colorimetric method and sodium nitrite-aluminum nitrate method,respectively.Extraction procedures of total polyphenols and total flavonoids from black chokeberry were optimized by orthogonal experiments.Antioxidant activities of different parts of black chokeberry were evaluated by DPPH and ABTS radical scavenging assays,hydroxyl radical scavenging rate,and reducing power assays.The results showed that total polyphenol content varied significantly among different parts of black chokeberry,ranked as leaves>fruits>stems>roots;total flavonoid content among different parts of black chokeberry also differed significantly,ranked as fruits>leaves>stems>roots.The optimal extraction conditions for total polyphenols were determined as extraction with 55%solution containing 0.2%hydrochloric acid at a material-to-liquid ratio of 1:25,ultrasonically extracted at 40℃for 65 minutes;for total flavonoids,extraction with 60%ethanol at a material-to-liquid ratio of 1:40,ultrasonically extracted at 65℃for 20 minutes was optimized.The optimized extraction methods for total polyphenols and flavonoids were found to be stable and feasible.All parts of black chokeberry exhibited strong antioxidant activities,with leaves and fruits showing higher effects comparable to vitamin C,while stems and roots showed lower activities than fruits.
