Abstract
To investigate the extraction process of multiple components from Gastrodia elata and its immune regulatory function,the sulfuric acid phenol extraction method and HPLC were used to analyze multiple components from Gastrodia elata.The single factor method was used to explore the factors such as extraction solvent,extraction time,and solid-liquid ratio on the extraction of multiple components,and the process was then optimized by orthogonal method.The optimal extraction conditions were obtained as:alcohol concentration of 10%,reaction temperature of 80℃,solid-liquid ratio of 1∶30,and reaction time of 50 minutes.,under which the extraction yield was 40.78%,the polysaccharide content in the extract was 59.97%,the gastrodin content was 0.52%,and p-hydroxybenzoic acid was 0.07%.Immunomodulatory functions were evaluated using a zebrafish model.The results showed that the maximum tolerated concentration(MTC)of Gastrodia elata extract in model zebrafish was 1000 μg/mL.The 95%extract of Gastrodia elata exhibited a promoting effect on macrophage phagocytosis at 62.5〜1000 μg/mL,demonstrating superior immune regulatory function compared with extracts from other parts of Gastrodia elata and positive controls.关键词
