Optimization of ultrasound-assisted cellulase extraction of polyphenols from Gastrodia elata and evaluation of their activity in vitro

Abstract

This study aimed to optimize the extraction process of Gastrodia elata polyphenols and evaluate their bioactivities in vitro.Fresh G.elata was used as the raw material.Single-factor experiments were first conducted to examine the key variables influencing polyphenol extraction efficiency.Subsequently,response surface methodology was applied to determine the optimal conditions for ultrasonic-assisted cellulase extraction.The optimized extraction parameters were:liquid-to-material ratio of 31:1,ethanol concentration of 73%,cellulase content of 1.3%,and ultrasonic power of 290 W,resulting in a polyphenol extraction rate of 0.62%.In vitro activity assays revealed that the half-maximal inhibitory concentrations(IC50)for ABTS+,DPPH,and hydroxyl radicals were 43.44,65.75,and 31.15 μg/mL,respectively.Regarding glucose-lowering activity,the IC50 values for α-amylase and α-glucosidase inhibition were 41.29 and 22.40 μg/mL,respectively.These findings provided a useful reference for the efficient extraction and in vitro activity evaluation of G.elata polyphenols using ultrasound-assisted cellulase methods.
