Abstract

The objective of the present study was to optimise an ultra performance liquid chromatography(UPLC)method for the determination of seven anthocyanins in grapes and wines.Following a rigorous comparison of the separation effects exhibited by the Shim-pack GIST C18 column(2.1 mm×100 mm,2 μm)and the Kinetex PFP column(3 mm×100 mm,2.7 μm),it was determined that the latter was to be selected as the stationary phase.The grape samples were extracted using formic acid:methanol(2∶98,V/V),and the wine samples were injected directly.The mobile phase A was composed of formic acid in water,with a volume ratio of 3∶97,while mobile phase B was a mixture of formic acid and acetonitrile,with a volume ratio of 2∶98.The separation process was conducted in gradient elution mode,with a detection wavelength of 518 nm and an injection volume of 1 μL.It was demonstrated that under optimised chromatographic conditions,the seven anthocyanosides were successfully separated within 11 minutes.Furthermore,an excellent linear relationship was observed between the concentration and the peak area(R2>0.9981).The range of recoveries was from 87.3%to 92.91%,with a precision(RSD)value below 1%.The method has been demonstrated to facilitate rapid and accurate qualitative and quantitative analysis of anthocyanosides in grapes and wines.It is characterised by high reliability and significant application value.
