Abstract

[Objective]To investigate the antioxidant effects and mechanisms of Phyllanthus emblica crude polysaccharides at different concentrations on the nematode using Caenorhabditis elegans as a model organism.[Method]UV-visible spectrophotometry was employed to measure the scavenging abilities of Phyllanthus emblica crude polysaccharides on DPPH,ABTS,and hydroxyl radicals.Enzyme assays were conducted to determine the activities of superoxide dismutase(SOD)and catalase(CAT)in C.elegans fed with 0,100,250,and 500 mg/L of Phyllanthus emblica crude polysaccharides.QPCR was used to assess changes in the mRNA levels of age-1,gpd-1,daf-16,sir-2.1,and clt-1 genes in C.elegans exposed to the same concentrations of Phyllanthus emblica crude polysaccharides.A high-oxygen environment was established to record the survival rates of C.elegans fed with 0,100,250,and 500 mg/L of Phyllanthus emblica crude polysaccharides and to observe the nuclear localization of DAF-16::GFP and changes in fluorescence intensity.[Result]Phyllanthus emblica crude polysaccharides exhibited strong scavenging abilities against DPPH,ABTS,and hydroxyl radicals.At concentrations of 100,250,and 500 mg/L,Phyllanthus emblica crude polysaccharides increased SOD activity in C.elegans,with 500 mg/L enhancing CAT activity.The polysaccharides also improved the survival rate of C.elegans under 1‰ H2O2-induced oxidative stress conditions.Additionally,the relative expres-sion levels of ctl-1,sir-2.1,and daf-16 genes in C.elegans were elevated to varying degrees,while the expres-sion of the age-1 gene showed a decreasing trend.Furthermore,there was no significant change in the nuclear localization of DAF-16::GFP,but fluorescence intensity was notably enhanced at 250 and 500 mg/L concentra-tions of Phyllanthus emblica Linn crude polysaccharides.[Conclusion]Phyllanthus emblica crude polysac-charides significantly enhanced SOD and CAT activities in C.elegans,improving their survival under high-oxygen stress conditions and conferring protective effects.Moreover,these polysaccharides upregulated the rela-tive expression of ctl-1,sir-2.1,and daf-16 genes while downregulating age-1 gene expression,suggesting that Phyllanthus emblica crude polysaccharides may modulate the antioxidant capacity of C.elegans through the DAF-16 signaling pathway.
