摘要

Abstract

[Objective]To study the immunomodulatory effect and mechanism of silver carp(Hypophthalmichthys molitrix)on immunosuppressed mice induced by cyclophosphamide and to evaluate its safety.[Method]Sixty mice were divided into 6 groups with 10 mice in each group:blank group,model group,ginseng control group(1 g ginseng formula granules/kg)and silver carp meat low,medium and high dose groups(1.6,3.3,6.6 g fish meat/kg).Except for the blank group,the other mice were injected with cyclophosphamide(80 mg/kg)to establish the immunosuppression model.After successful modeling,the weight,blood routine and other indicators of mice were detected,combined with metabonomics technology to analyze the mechanism,and the acute toxicity test was carried out to evaluate the safety.[Result]Compared with the model group,the body weight(P<0.05),thymus index(P<0.01),white blood cell(WBC),hemoglobin(HGB)and platelet(PLT)counts in the silver carp meat middle dose group were significantly increased(P<0.05).The concentra-tion ratio of cyclic adenosine monophosphate(cAMP)/cyclic guanosine monophosphate(cGMP)(P<0.05)and ATPase activity(P<0.01)were increased,and the contents of immunoglobulin G(IgG)and immunoglobulin M(IgM)were significantly up-regulated(P<0.001).Metabonomics analysis showed that the differential metabolites were mainly enriched in amino acid and lipid metabolic pathways,and played a role by regulating D-amino acid metabolism and other pathways,and there was no obvious toxicity in acute toxicity test.[Conclu-sion]Silver carp meat can improve the hypoimmunity by regulating the function of immune organs,repairing the energy metabolic network and regulating the level of metabolites,and it is safe within the experimental dose.
