Abstract

To enhance the polysaccharide extraction yield from sarcodon aspratus and investigate its in vitro bioactivity and monosaccharide composition,a method combining ionic liquids with ultrasound-assisted extraction was optimized.Using sarcodon aspratus as the raw material and the polysaccharide extraction yield as the evaluation index,single-factor and response surface methodology(RSM)were employed to optimize the extraction process.The in vitro bioactivity and polysaccharide composition were also studied.The results showed that 1-ethyl-3-methylimidazolium bromide was the optimal ionic liquid.The optimal extraction parameters were determined as follows:solid-to-liquid ratio of 1∶30(g/mL),ionic liquid volume of 1.5 mL,ultrasound temperature of 44℃,and ultrasound time of 36 min.Under these conditions,the polysaccharide extraction yield reached 42.54%.The polysaccharide was composed of mannose,rhamnose,glucuronic acid,galacturonic acid,glucose,galactose,arabinose,and fucose,with a mass ratio of 0.98∶0.33∶0.12∶0.79∶29.16∶4.69∶0.25∶8.93.At a concentration of 1 mg/mL,the scavenging rates of the polysaccharide against DPPH•,•OH,and ABTS+• were 85.65%,75.69%,and 76.88%,respectively,while its inhibition rates on α-amylase and pancreatic lipase were 75.1%and 78.8%,respectively.This research provides a reference for the extraction of active substances using ultrasound-assisted ionic liquid methods and for the application of sarcodon aspratus polysaccharides in functional foods.
