超声辅助离子液体法提取虎掌菌多糖及其体外活性、单糖成分研究
Study on the extraction of sarcodon aspratus polysaccharides by ultrasound-assisted ionic liquid method and their in vitro activity and monosaccharide composition

Abstract
To enhance the polysaccharide extraction yield from sarcodon aspratus and investigate its in vitro bioactivity and monosaccharide composition,a method combining ionic liquids with ultrasound-assisted extraction was optimized.Using sarcodon aspratus as the raw material and the polysaccharide extraction yield as the evaluation index,single-factor and response surface methodology(RSM)were employed to optimize the extraction process.The in vitro bioactivity and polysaccharide composition were also studied.The results showed that 1-ethyl-3-methylimidazolium bromide was the optimal ionic liquid.The optimal extraction parameters were determined as follows:solid-to-liquid ratio of 1∶30(g/mL),ionic liquid volume of 1.5 mL,ultrasound temperature of 44℃,and ultrasound time of 36 min.Under these conditions,the polysaccharide extraction yield reached 42.54%.The polysaccharide was composed of mannose,rhamnose,glucuronic acid,galacturonic acid,glucose,galactose,arabinose,and fucose,with a mass ratio of 0.98∶0.33∶0.12∶0.79∶29.16∶4.69∶0.25∶8.93.At a concentration of 1 mg/mL,the scavenging rates of the polysaccharide against DPPH•,•OH,and ABTS+• were 85.65%,75.69%,and 76.88%,respectively,while its inhibition rates on α-amylase and pancreatic lipase were 75.1%and 78.8%,respectively.This research provides a reference for the extraction of active substances using ultrasound-assisted ionic liquid methods and for the application of sarcodon aspratus polysaccharides in functional foods.关键词
