Abstract

The use of salt-tolerant rootstocks is one of the effective technical approaches to enhance the salt tolerance of grapevines.In this study,'Cabernet Sauvignon/A15'grafted seedlings were treated with 100 mmol·L-1 NaCl(with water as the control).Based on intervarietal single nucleotide polymorphism(SNP)sites,the characteristics of mRNA transport between rootstock and scion were identified and analyzed.Through screening for long distance transport potential,a total of 490 transportable mRNAs were identified.Under control conditions,383 mRNAs were detected.Under salt stress,337 transportable mRNAs were detected.230 were detected simultaneously in both salt treatment and control.The length of transportable mRNAs ranged from 138 to 6982 bp,with no significant correlation observed between transport frequency and mRNA length.GO functional analysis indicated that under salt stress,transportable genes were primarily enriched in functional categories such as binding,metabolic processes,and cell periphery.KEGG analysis revealed that plant hormone signal transduction was the most significantly enriched pathway under salt stress,with a notable increase in the number of differentially expressed genes,indicating enhanced enrichment under stress.Furthermore,salt stress increased the transport level of some genes.This study systematically identified transportable mRNAs between rootstock and scion and evaluated the effects of salt stress on their transport characteristics,providing a theoretical basis for elucidating the molecular mechanisms underlying rootstockscion interactions in improving salt tolerance in grapevines..
