Abstract

In order to clarify the influence of nutritional regulation on grape skin coloring and related gene expression,the'Red Barado'grape was selected as the material,an interactive experiment was conducted among 4 basic fertilizers(A:pig manure,B:1/3 cow manure+1/3 sheep manure+1/3 pig manure,C:2/3 cow manure+1/3 sheep manure,D:1/2 cow manure+1/2 sheep manure),and 2 formulations of water-soluble fertilizer(formulations 1:10N,30P,20K and 2:Biojing potassium water-soluble fertilizer and Anhong).Pig manure without water-soluble fertilizer was used as control treatment(CK).The anthocyanin contents,the activity and the relative gene-expression levels of related enzymes were used to determine in grape skin of'Red Barado'at the maturity stage.The results showed that compared with other treatments,the coloring of grape skins was the best under the combined treatment of 1/2 cow manure+1/2 sheep manure as base fertilizer and formula 2 water-soluble fertilizer.The solid-acid ratio of the fruit was 2.96 times that of the control,total phenols(57.49 μmol·L-1),flavonoids(7.08 mg·g-1),and anthocyanins(7.77 μmol·g-1)of the fruits were all the highest.At the same time,the activities of enzymes related to anthocyanin synthesis in fruit peels(F3'H,F3'5'H,LDOX and UFGT)were significantly enhanced,and the relative expression levels of related genes(PAL,F3'H,F3'5'H,DFR)were the highest.The results show that the combined treatment of 1/2 cow manure+1/2 sheep manure as base fertilizer and formula 2 water-soluble fertilizer has the best effect on promoting the coloring of the skins of'Red Barado'grapes.
