Abstract
This study optimized the extraction process of Magnoliae flos polysaccharides(MFP)using an environmentally friendly microwave-assisted deep eutectic solvent(DES)technology,and systematically evaluated their antioxidant and antifatigue activities.Single-factor experiments combined with Box-Behnken response surface methodology was employed to determine the optimal extraction conditions.The optimal extraction conditions were determined as follows:a choline chloride-1,4-butanediol(1∶1)based DES system(with a water content of 61.40%),a liquid-to-material ratio of 41.2 mL/g,an extraction temperature of 103.6℃,and an extraction time of 16.0 min.Under these conditions,the polysaccharide yield reached 111.8±4.3 mg/g,representing a 38.6%increase compared with conventional extraction methods.Animal experiments demonstrated that administration of 400 mg/kg MFP significantly prolonged the exhaustive swimming time of mice by 43.3%,reduced mice serum urea nitrogen and blood lactic acid levels by 27.9%and 29.0%,respectively,and increased mice hepatic and muscle glycogen contents by 42.6%and 86.7%,respectively.Antioxidant activity analysis further revealed that the high-dose MFP group mice hepatic malondialdehyde levels were markedly decreased by 25.4%,while mice superoxide dismutase and lactate dehydrogenase activities were enhanced by 54.3%and 76.2%,respectively.This study was the first to demonstrate that MFP exerted antifatigue effects through the regulation of energy metabolism and enhancement of the antioxidant defense system.The optimized DES-microwave synergistic extraction process for MFP provided a promising and sustainable approach for the high-value utilization of plant polysaccharides.关键词
