Abstract

This study investigated the protective effects and underlying mechanisms of sesamin against acetaminophen(APAP)-induced acute liver injury in mice.Forty C57BL/6 mice were randomly divided into control group,model group,low-dose sesamin group(100 mg/kg),and high-dose sesamin group(200 mg/kg).Following 21 days of oral gavage,acute liver injury was induced via intraperitoneal injection of APAP(300 mg/kg).Pathological changes in mice liver tissue were evaluated using HE staining.Serum ALT and AST activities,liver MDA content,and SOD activity were measured in mice.Additionally,inflammatory factors IL-1β,IL-6,and TNF-α levels were detected by ELISA,and protein expression of Nrf2,HO-1,and NQO1 were analyzed by Western blot.The results showed that compared with the model group,sesamin treatment significantly alleviated mice liver tissue pathological damage,reduced serum transaminase levels,MDA content,and inflammatory factor expression,and increased SOD activity.Furthermore,sesamin dose-dependently increased SOD activity and upregulated the expression of Nrf2,HO-1,and NQO1 in mice.The results indicated that sesamin could enhance antioxidant capacity and reduce inflammatory response by activating the Nrf2/HO-1/NQO1 pathway,thereby effectively alleviating APAP-induced liver injury in mice.This study provides a theoretical basis for the development of sesamin as a potential hepatoprotective agent.
