Effects of exogenous selenium combined with rhizobium treatment on growth and active components of Astragalus membranaceus var.mongholicus

通过外源硒与根瘤菌处理后,采用发芽床试验法测定蒙古黄芪种子的萌发指标,采用土培盆栽法测定蒙古黄芪幼苗生长指标、抗氧化酶活性和活性成分含量.结果显示外源硒、根瘤菌和外源硒结合根瘤菌 3种处理对蒙古黄芪种子的萌发均有显著的抑制作用(P<0.05),对幼苗的生长均有促进作用;外源硒结合根瘤菌处理,蒙古黄芪幼苗SOD活性、POD活性、CAT活性分别增加了 29.00%、27.90%、46.31%,与空白处理相比均有显著增加(P<0.05);外源硒与根瘤菌结合处理,蒙古黄芪中粗脂肪、硝态氮、总皂苷含量均最高,分别为 4.77%、1.54%、0.51%.外源硒结合根瘤菌对蒙古黄芪幼苗抗氧化酶活性、粗脂肪、硝态氮、总皂苷含量有明显的促进作用.该研究可为药食两用富硒蒙古黄芪的生产和品质的提高奠定基础并提供技术参考.

After treatment with exogenous selenium and rhizobium,the germination index of Astragalus membranaceus var.mongholicus(AMM)seeds was determined by germination bed test,and the growth index,antioxidant enzyme activity and active component content of AMM seedlings were determined by soil culture pot method.The results showed that exogenous selenium,rhizobium and exogenous selenium combined with rhizobium had significant inhibitory effects on the germination of AMM seeds(P<0.05),but it promoted the growth of seedlings.Using exogenous selenium combined with rhizobium treatment,the SOD activity,POD activity and CAT activity of AMM seedlings increased 29.00%,27.90%and 46.31%,respectively,with significantly increase compared with the blank treatment(P<0.05).The content of crude fat,nitrate nitrogen and total saponins in AMM were the highest with 4.77%,1.54%and 0.51%respectively in the combination of exogenous selenium and rhizobium treatment.Exogenous selenium combined with rhizobium significantly promoted antioxidant enzyme activity,crude fat,nitrate nitrogen and total saponin content of AMM seedlings.This study can lay a foundation and provide technical reference for the production and quality improvement of selenium-enriched Mongolian AMM for both medicine and food usage.
