Optimization of extraction process of polysaccharide from artificially cultivated Sanghuangporus baumii and its antioxidant activity

以超声辅助热水浸提法提取人工栽培暴马桑黄多糖,采用单因素实验考察料液比、超声时间、浸提时间的影响,并用Box-Behnken响应面试验优化提取工艺,测定暴马桑黄多糖FRAP总还原力,对羟基自由基、ABTS+自由基、DPPH自由基的清除能力,评价其抗氧化能力;结合形态学观察和MTT法考察暴马桑黄多糖对H2O2 诱导PC12 细胞模型的保护作用.单因素和响应面实验结果表明,当提取条件为料液比1∶30 g/mL,超声时间 15 min,浸提时间 60 min时,人工栽培暴马桑黄多糖提取率最高为1.31%.对DPPH、OH和ABTS+自由基的IC50 值分别为 0.403,2.870 和 0.142 mg/mL,表明暴马桑黄多糖具有良好的抗氧化能力,细胞实验证明暴马桑黄多糖对由H2O2 建立的氧化损伤模型有保护作用.说明暴马桑黄多糖具有较强的抗氧化能力和抑制细胞氧化损伤的作用.

Ultrasound-assisted hot water extraction was used to extract the polysaccharides of Sanghuangporus baumii.The effects of solid-liquid ratio,ultrasound time,and extraction time on the extraction rate were studied by single factor experiment and optimized by Box-Behnken response surface method.The total reducing power of FRAP,the scavenging capacity of hydroxyl,ABTS and DPPH radicals were determined to evaluate the antioxidant capacity of polysaccharides from Sanghuangporus baumii.The antioxidative effects of polysaccharides from Sanghuangporus baumii were examined by H2O2-induced PC12 cell model,where morphological observation and MTT assay were performed.The results of single factor test and response surface experiments showed that the highest extraction rate of 1.31%of polysaccharides was achieved at the extraction conditions of 1∶30 g/mL solid-liquid ratio,15 min ultrasound,and 60 min extraction time.The IC50 values for DPPH·,·OH and ABTS+ radicals were 0.403,2.870 and 0.142 mg/mL,respectively,which indicated that the polysaccharides from Sanghuangporus baumii had good antioxidant capacity,and cell experiments proved that the polysaccharide from Sanghuangporus baumii had a protective effect on the oxidative damage model established by H2O2,this indicated that the polysaccharides from Sanghuangporus baumii had strong antioxidant capacity and inhibitory effects on cellular oxidative damage.
