检测库尔勒香梨中性多糖(Pyrus sinkiangensis Yu.neutral polysaccharide,PSNP-1、PSNP-2)的结构、抗氧化活性以及对过敏性哮喘小鼠的影响和机制.利用柱色谱分离得到了PSNP-1、PSNP-2两组分.PSNP-1、PSNP-2 的总糖含量分别为 91.81%、98.62%,糖醛酸含量分别为 2.94%、3.11%,分子量分别为 1503.14 kDa、26.73 kDa,其中PSNP-1 的单糖组成及摩尔比主要为葡萄糖∶半乳糖∶阿拉伯糖=54.6∶5.1∶2.9 及少量甘露糖和鼠李糖,PSNP-2 主要为甘露糖∶葡萄糖∶半乳糖∶阿拉伯糖=1.8∶38∶10.5∶33 及少量的鼠李糖.两种多糖均有多羟基糖的特征吸收峰并且是以α-糖苷键和β-糖苷键相连的吡喃糖环中性多糖且均有三股螺旋结构.体外抗氧化实验显示PSNP-1、PSNP-2 有良好的抗氧化活性,且对OVA诱导的哮喘小鼠有改善作用,其作用机制可能通过干预 TLR4/NF-κB 信号通路从而改善 OVA 诱导的哮喘小鼠气道炎症.

The structure and antioxidant activity of neutral polysaccharide(PSNP-1,PSNP-2)from Pyrus sinkiangensis Yu were detected,and effect and effect mechanism of neutral polysaccharide on mice with allergic asthma were investigated.Two components PSNP-1 and PSNP-2 were obtained from Pyrus sinkiangensis Yu by column chromatography.The total sugar content of PSNP-1 and PSNP-2 were 91.81%and 98.62%,respectively.Their uronic acid content was 2.94%and 3.11%,and their molecular weight was 1503.14 and 26.73 kDa,respectively.The monosaccharide composition of PSNP-1 were glucose,galactose,and arabinose with a ratio of 54.6∶5.1∶2.9,and a small amount of mannose and rhamnose.PSNP-2 was mainly composed of mannose,glucose,galactose and arabinose,with a ratio of 1.8∶38∶10.5∶33,and small amount of rhamnose.Both polysaccharides had absorption peaks of polyhydroxy sugars and neutral pyranose rings were connected by α-glucoside and β-glucoside bonds.In vitro antioxidant experiments showed that PSNP-1 and PSNP-2 had good antioxidant activities and could improv symptoms of airway inflammation of OVA-induced asthma in mice.The mechanism may be through the intervention of TLR4/NF-κB signaling pathway to improve the airway inflammation of OVA-induced asthma.
