Grafting ferulic acid onto xylo-oligosaccharide from walnut shells and analysis of its antioxidant activity

为提高核桃果壳的经济价值,本研究以其为原料制备阿魏酸接枝化的低聚木糖.首先以核桃皮粉为原料提取木聚糖;在无氧条件下,对木聚糖进行阿魏酸接枝化;再利用木聚糖酶对阿魏酸木聚糖(FXL)进行酶解得到阿魏酸低聚木糖(FXO);在制备过程中分别采用紫外可见分光光度计(UV-Vis)、傅里叶变换红外光谱(FT-IR)、核磁共振(NMR)分别对产物进行鉴定;最后将FXO用于抗氧化的研究.紫外可见分析结果表明,木聚糖中核酸与蛋白质已经被去除干净;通过FT-IR分析表明,已经合成了FXL;经过氢谱与碳谱分析,获得了FXO.抗氧化结果显示,木聚糖、FXL和FXO均表现出抗氧化活性;并且,阿魏酸接枝化能够提高多糖的抗氧化性能.相比木聚糖、阿魏酸、FXL,FXO的抗氧化活性与还原能力最强.本研究为核桃皮的利用提供了一定的研究基础.

In order to enhance the economic value of walnut shells,using walnut shells as raw material to prepare xylo-oligosaccharides grafted with ferulic acid.Firstly,xylan was extracted from walnut shell powder.Under anaerobic conditions,xylan was grafted with ferulic acid.Ferulic acid oligomeric xylan(FXO)was obtained by enzymatic hydrolysis of the ferulic acid xylan(FXL)using xylanase.During the preparation process,UV-Vis,FT-IR,and NMR were used for identification of the products.FXO was then used for antioxidant research.UV-Vis analysis results indicated that the nucleic acid and protein had been effectively removed from the xylan;FT-IR analysis confirmed that FXL had been synthesized.while analysis of proton and carbon spectra led to the isolation of FXO.Antioxidant results showed that xylan,FXL,and FXO all exhibited antioxidant activity,and the grafting of ferulic acid enhanced the antioxidant properties of polysaccharides.Compared to xylan,ferulic acid and ferulic acid xylan,FXO showed the strongest antioxidant activity and reduction capability.This study provides a foundation for the utilization of walnut shells.
