北沙参提取物改善阴虚便秘作用研究OACSTPCD

Effects of Glehniae radix aqueous extract on improvement of Yin-deficiency and constipation

本研究旨在探索北沙参提取物改善便秘的效应及机制,探索不同加工方式的差异.采用UHPLC-HRMS法测定北沙参水提物主要成分;采用洛哌丁胺和左甲状腺素钠建立阴虚便秘大鼠模型后,灌胃给予两种加工方式北沙参水提物(生药量 1 g/kg)7 天,检测大鼠代谢指标,观察结肠组织形态变化;阿利新蓝染色法及免疫组织化学法观察结肠组织中粘液分泌情况及 5-羟色胺 4 受体(5-HT4R)的表达.结果表明:未去皮水提物较去皮水提物中成分更为丰富;两种提取物给药干预后,第一黑便时间和小肠推进率均显著改善(P<0.05),结肠黏蛋白、5-HT4R显著增加(P<0.05).本研究阐明北沙参水提物通过增加结肠黏液分泌,修复肠道黏液屏障,调节5-HT途径,兴奋肠道平滑肌,从而实现缓解便秘的作用.

In order to explore the effects and mechanisms of Glehniae radix aqueous extract in improving constipation and to explore the differences in different processing methods.The main components of the Glehniae radix aqueous were determined by UHPLC-HRMS method;After establishing a Yin deficiency constipation rat model with loperamide and levothyroxine sodium,mice were orally administered with two processed forms of Glehniae radix aqueous extract(1 g/kg of crude herbs)for 7 days.Metabolic indicators of rats were measured,and changes in colonic tissue morphology were observed;Alcian blue staining and immunohistochemical method was used to observe mucus secretion and detect the protein expression of 5-HT4R of mice colon tissues.The results showed that the unpeeled aqueous extract was richer in constituents than the peeled aqueous extract.After treatment with the aqueous extract of Glehniaer radix,the time of the first black stool and the rate of small intestinal propulsion were significantly improved(P<0.05),mucin and the content of 5-HT4R in the colonic tissues was significantly increased(P<0.05).The study elucidated that the Glehniae radix aqueous extract relieved Yin-deficiency and constipation by increasing colonic mucus secretion and repairing intestinal mucus barrier,and regulating the 5-HT pathway to stimulate intestinal smooth muscle.
