本文建立了电位滴定测定面粉添加剂葡萄糖氧化酶活度的新方法.葡萄糖氧化酶催化氧化葡萄糖产生H2O2,高价铈(Ce4+)与 H2O2 发生 2∶1 的定量反应,当用Ce4+标准溶液滴定H2O2 时,H2O2 生成量直接体现了葡萄糖氧化酶活度,考察了滴定条件,系统分析了方法的准确度和精密性.结果表明,葡萄糖氧化酶活度在0.00~3.40 U/mL范围内与Ce4+标准溶液滴定体积呈线性关系:∆V(Ce4+)=6.061X(U/mL)-0.0223,相关系数为 0.9988,方法的最低检出量为 4.94×10-3 U/mL.自配葡萄糖氧化酶工作液作测试液,平行 6 次测定,相对标准偏差RSD<5%.

A new method for the potentiometric titration determination of glucose oxidase activity in flour additives was established.Glucose oxidase catalyzes the oxidation of glucose to produce H2O2,Cerium(IV)ion(Ce4+)undergone a 2∶1 quantitative reaction with H2O2,where when titrating H2O2 with a standard Ce4+solution,the amount of H2O2 generated directly reflected the glucose oxidase activity.The titration conditions were investigated,and the accuracy and precision of the method were systematically analyzed.The results showed that the glucose oxidase activity in the range of 0.00 to 3.40 U/mL was linearly related to the titration volume of the standard Ce4+solution:∆V(Ce4+)=6.061X(U/mL)-0.0223,with a correlation coefficient of 0.9988.The method's limit of detection was 4.94×10-3 U/mL.Using a self-prepared glucose oxidase working solution as the test solution,six parallel measurements were conducted with a relative standard deviation(RSD)of less than 5%.
