高效液相色谱法测定非变性Ⅱ型胶原蛋白含量的研究OACSTPCD
Determination of non-denatured type Ⅱ collagen content by high performance liquid chromatography
研究非变性Ⅱ型胶原蛋白含量的测定方法,对样品中的目标成分进行测定和方法学验证分析.通过高效液相色谱法(HPLC)测定非变性Ⅱ型胶原蛋白的含量,考察线性、精密度、准确度、耐用性.实验结果表明,非变性Ⅱ型胶原蛋白(以L-羟脯氨酸计),在 0～398.48 μg/mL范围内线性关系较好;精密度较好;平均回收率为99.2%,RSD为0.87%;耐用性较好.经过全面的方法学验证,该实验方法稳定可靠,适用于高效液相色谱法测定非变性Ⅱ型胶原蛋白的含量,并可应用于相关产品的质量控制工作.
A method for the determination of non-denatured type Ⅱ collagen content was investigated for the determination of the target components in the samples and methodological validation of the analysis.The linearity,precision,accuracy and durability were investigated by high performance liquid chromatography(HPLC)for the determination of non-denatured type Ⅱ collagen content.The results showed that the non-denatured type Ⅱ collagen(as L-hydroxyproline)had good linearity in the range of 0～398.48 μg/mL;good precision;mean recovery of 99.2%with RSD of 0.87%;and good durability.After comprehensive methodological validation,the experimental method was stable and reliable and was suitable for the determination of non-denatured type Ⅱ collagen content by high performance liquid chromatography and could be applied to the quality control work of related products.
