大蝎子草总多酚提取工艺优化及抗氧化活性OA北大核心CSTPCD

Optimization of Extraction Process and Antioxidant Activity of Total Polyphenols from Girardinia diversifolia

本试验采用超声辅助乙醇提取大蝎子草地上部分总多酚,通过单因素试验结果设计Box-Behnken模型,优化大蝎子草地上部分总多酚最佳工艺条件,用DPPH自由基清除率、FRAP法评价大蝎子草地上部分总多酚的抗氧化性.结果显示,乙醇浓度、液固比、超声时间、超声温度的最佳工艺参数分别为75%、50 mL/g、35min、40 ℃时,总多酚提取率为2.81%,与Box-Behnken模型的预测值相符.抗氧化性试验表明,0.15 mg/mL大蝎子草总多酚提取液DPPH清除率可达75.8%,半数抑制浓度IC50值为0.057 mg/mL,具有一定的抗氧化性,为大蝎子草野生资源的研究和利用提供相应的理论支撑,为新型饲料添加剂的开发提供理论基础.

The ultrasonic-assisted ethanol was used to extract the total polyphenols from the aerial part of Girardinia diversifolia in this experiment.Box-Behnken model was designed based on the results of single-factor experiments to optimize the optimal process conditions of total polyphenols from the aerial part of Girardinia diversifolia.DPPH free radical scavenging rate and FRAP method were used to evalu-ate the antioxidant capacity of total polyphenols from the aerial part of Girardinia diversifolia.Results showed when the optimal process parameters as ethanol concentration,liquid-solid ratio,ultrasonic time and ultrasonic temperature were 75%,50 mL/g,35 min and 40 ℃,respectively,the extraction rate of to-tal polyphenols was 2.81%,which was consistent with the predicted value of Box-Behnken model.The an-tioxidant test showed that the DPPH radical-scavenging activity of 0.15 mg/mL total polyphenol from the aerial part of Girardinia diversifolia could reach 75.8%,and the IC50 value of the half inhibitory concen-tration was 0.057 mg/mL,which had certain antioxidant capacity.It provides corresponding theoretical da-ta support for further research and utilization of wild resources of Girardinia diversifolia,and a theoreti-cal basis for the development of new feed additives.
